Emergency services were called to the scene just before 4pm on Saturday.
The woman was brought to a boat ramp, where members of the public tried to revive her with CPR, but she was pronounced dead at the scene.
The boat's driver, a man in his 20s, was not injured in the incident, and will undergo mandatory blood and urine tests.
A second man onboard the boat at the time was also uninjured.
It's the second death on Lake Conjola this summer, after a Korean man drowned while snorkelling on Christmas Day.
South coast police have launched an investigation into Saturday's incident and are appealing for any witnesses to come forward.
A report will be prepared for the Coroner.
