After racing to 1-98 overnight, India batted in more dogged fashion in the morning session at Mumbai on Friday to trim Australia's lead to 26.
Smriti Mandhana (74) looked in imperious touch before being run out, but her exit proved only temporary relief for Australia's bowlers, with 20-year-old debutant Richa Ghosh (24no) and Jemimah Rodrigues (24no) unbeaten at lunch.
Nightwatcher Sneh Rana (9) frustrated the tourists' attack for 57 balls before being bowled by Ash Gardner.
Mandhana appeared a lock for her second Test century against Australia before running too casually when called through for a sharp single by Ghosh and caught fractionally short by Kim Garth's throw to Gardner.
Ghosh shrugged off the disappointment to play some nice strokes, but she should have been gone for 14 when Ellyse Perry's nightmare match - she was dismissed for four on the opening morning before bowling four overs for 31 - continued when she dropped a sitter at mid-off.
India's batters are continuing the fine job of their bowlers, who knocked Australia over for 219 on the first day.
New vice-captain Tahlia McGrath (50 off 56 balls), captain Alyssa Healy (38) and tail-ender Garth (28no) were the only Australian batters to impress against a potent Indian attack led by seamer Pooja Vastrakar, who took a career-best 4-53.
